Optimized Library Construction for High-quality Transcriptome Sequencing on the Illumina® Platform

Kapa Biosystems is proud to announce our very own Dr. Adriana Geldart for a special webinar on how to overcome the numerous challenges associated with library preparation for transcriptome sequencing. High-quality library preparation is key in transcriptome sequencing to ensure that the data acquired at the end of sequencing is accurate. Dr. Geldart will present data on the ideal quality control metrics to use when preparing RNA-Seq libraries, and show that these metrics assist in predicting the outcome of the sequencing run.

Abstract

Transcriptome sequencing is an evolving and challenging field of molecular biology.  During product development, Kapa Biosystems has addressed many of the challenges associated with acquiring the highest quality transcriptome data including library amplification bias and coverage of difficult regions. The resultant KAPA Stranded mRNA-Seq Kit offers a streamlined workflow and exhibits precise measurement of strand orientation and uniform coverage, and high confidence mapping of alternate transcripts, including GC-rich and low abundance transcripts.

In collaboration with Perkin Elmer, we have successfully developed automation scripts for the KAPA Stranded mRNA-Seq Kit on the Caliper Sciclone NGS instrument, and have profiled Mcf7 breast cancer cell lines with a key opinion leader in the field.  We will present on the quality control metrics that were utilized during development and how they are indicative of library outcome and overall coverage.  Lastly, the data presented will analyze key sequencing metrics for RNA input titrations and reproducibility between samples.

About Dr. Adriana Geldart

Dr. Adriana Geldart started her scientific education at Stonehill College with a Bachelor’s degree in Biology. She completed her PhD degree at UMass Medical School in Biomedical Sciences, where she focused on the analysis of long-range chromosomal interactions in Saccharomyces cerevisiae.

Dr. Geldart went on to a postdoctoral fellowship at Brigham & Women’s Hospital in conjunction with Harvard Medical School, where she investigated the effect of extracellular acidosis on the metabolic phenotype of pulmonary artery smooth muscle cells in relation to pulmonary hypertension. She then joined Ion Torrent as a Research and Development Scientist where she contributed to multiple projects, including the development of the OneTouch 2 automated sample preparation instrument.

Dr. Geldart joined Kapa Biosystems in 2013 and is a Support and Applications Manager.  Adriana is directly involved in supporting customers for our entire NGS product line, as well as collaborating externally to develop new applications.
